چکیده

Polymethylated Flavonols, Identification, Enzymatic Synthesis, O-Methyltransferase, Chrysosplenium americanum, Saxifragaceae The major flavonoid constituents of Chrysosplenium americanum (Saxifragaceae) were identi fied by UV-, NMRand mass spectroscopy and the analysis of their hydrolytic products. They consisted of two 2'-0-/?-D-glucosides of partially methylated 2'-hydroxyquercetin and four 5'-0-/?-D-glucosides of partially methylated 6-hydroxyquercetin (quercetagetin). The O-methyltransferase system of this tissue can catalyse the stepwise O-methylation of quercetin, but not quercetagetin, to its 3,7,4'-triOMe-dervative. It is postulated that the latter compound may serve as a branching-point intermediate in the biosynthesis of the naturally occurring O-methylated flavonols of this tissue.

Role of Serine 286 in cosubstrate binding and catalysis of a flavonol O-methyltransferase.

O-Methyltransferases catalyze the transfer of the methyl groups of S-adenosyl-L-methionine to specific hydroxyl groups of several classes of flavonoid compounds. Of the several cDNA clones isolated from a Chrysosplenium americanum library, FOMT3' encodes the 3'/5'-O-methylation of partially methylated flavonols. The recombinant protein of another clone, FOMTx which differs from FOMT3' by a sing...

Purification and Properties of Flavonol - Ring B

A novel glucosyltransferase which catalyzed the transfer of glucose from UDP-glucose to positions 2' and 5' of partially methylated flavonols was isolated from the shoots of Chrysosplenium americanum Schwein ex Hooker. It was purified 225-fold by ammonium sulfate precipitation and successive chromatography on Sephadex G-100, hydroxyapatite, and polybuffer ion exchanger. This glucosyltransferase...

Cell Wall-Bound Phenolics from Norway Spruce (Picea abies) Needles

Secondary products, such as phenolics, accumulate predominantly in the form of hydrophilic conjugates in the vacuoles (intraprotoplasmic storage) of plants [1—3]. Also there are many reports on their extraprotoplasmic location, e.g. as exudates [4] or cell wall components. The latter include the polymer lignin [5], as well as monomeric hydrolyzable phenolics, which are mainly hydroxycinnamic ac...

Triterpenoids of Chrysosplenium carnosum.

A phytochemical study of the ethanolic extract of Chrysosplenium carnosum Hook. f. et Thoms. led to the isolation of two new oleanane-type triterpenoids, 6β-hydroxy-3-oxoolean-12-en-27-oic acid (1) and 3β, 21α-dihydroxyolean-12-en-27-oic acid (2), along with fourteen known compounds (3-16), all of which were isolated from this plant for the first time. The structures of these compounds were est...

Influence of Flavonols As in vitro on Low Density Lipoprotein Glycation

The non-enzymatic glycation of Low density lipoprotein (LDL) is a naturally occurring chemical modification of apolipoprotein B as a result of condensation between lysine residues and glucose. Glycated LDL is poorly recognized by LDL receptors and initiates different processes that can be considered proatherogenic. Thus, LDL glycation may contribute in the increased atherosclerotic risk of pati...
